Controlling perfusion bioreactors

Simple control of perfusion on cell culture biorectors, based on infrequent daily sampling and estimation of the live cell concentration, will lead to large process deviations. A robust automatic perfusion rate control system based on the on-line Aber Biomass Monitor probe is now being used in cell culture manufacturing processes. Many of these are operating under cGMP conditions allowing the process to be operated under optimum conditions for maximum production of recombinant proteins or monocalonal antibodies. In the control algorithm, a cell specific perfusion rate is specified and the Biomass Monitor "live"cell density signal is averaged and converted into a perfusion flow rate.
